红平菇漆酶基因异源表达及对染料脱色的研究

Heterologous expression of Pleurotus djamor laccase gene in Pichia pastoris and decolorization on dyes by recombinant laccase

摘要: 以白腐菌红平菇Pleurotus djamor RNA为模板,通过RT-PCR得到漆酶Pd-lac1基因的完整CDS序列。构建毕赤酵母表达载体pPICZB-Pd-Lac1,并通过电转化法导入Pichia pastoris中,通过PCR结果证明,Pd-lac1的CDS序列部分阳性转化子已经整合到甲醇毕赤酵母染色体上,经摇瓶液体培养后筛选出蛋白表达较高的酵母工程菌株。漆酶活力达54 U/L。进而研究了重组漆酶对染料脱色的影响。结果表明:重组漆酶对蒽醌类SN4R脱色可达52.8%;杂环类中性红、偶氮类甲基橙和三苯基甲烷类孔雀绿的脱色率低于SN4R,脱色率分别为14.8%、14.5%、0.24%,显示出重组漆酶对SN4R脱色效率具有较大的应用潜力,从而对废水处理具有更广的应用前景。

Abstract: Regarding the White Rot Fungus Pleurotus djamor as the templet,the complete sequence of Laccase Gf-lac1's gene can be got through the RT-PCR. In this way,the expression vector of Pichia pastoris named pPICZB-Gf-Lac1 is built and it is imported into Pichia pastoris by electrotransformation .What's more,part of masculine transformants called PCR turns out that the sequence CDS of Gf-Lac1 has been integrated in the Pichia pastoris chromosome.Besides,after training in the shake-flask liquid, the protein with higher pastoris engineering strains are selected. Furthermore, the vitality of laccase is reached 53U/L, and the study that laccase restructured is influenced dyeware bleaching has been researched further. The result turns out that the bleaching rate -restructring laccase to Alizarin Red Anthraquinone can be reached 53.6%; on the other hand, the bleaching rates of Heterocyclic Neutral Red, Methyl Orange Azo and Triphenylmethane Malachite Green are lower than Alizarin Red, which are namely 15.5%, 15.1% and 0.23%. In the words, it shows that the efficiency of restrucruring laccase to Alizarin Red possesses larger potential applications, thus it owns wider applied prospects in wastewater treatment.
