Screening and applications of superior strain for biological retting of wild ramie

Abstract:

In order to achieve bio-degumming of the wild ramie, 37 strains were isolated from retting water of the wild ramie by agar plate dilution method, and superior stain was screened by transparent circle in combination with DNS method. It was found that transparent circle and enzymatic activity of TJ03 strain were the best. The ratio of transparent circle area and colony area was 19.98,pectinase activity and mannanase activity were 9.54, 8.63U/mL, respectively. Then microbial strain TJ03 was used for degumming of the wild ramie. The optimal conditions of retting wild ramie were pH 7.5, temperature 35℃, bath ratio 1:25 and 15 % inoculum size of strain TJ03, with residual gum content of wild ramie only 14.09%. Fiber bast was separated from the natural gum and woody matter of the stalk of wild ramie.
